1 硬件操作
熒光分光光度計(jì)在使用時(shí),需要注意開機(jī)次序,以保護(hù)設(shè)備之間不受影響。熒光光譜儀開機(jī)順序一般為先開氙燈,然后開儀器主機(jī),最后開跟儀器連接的計(jì)算機(jī)。如此操作的原因在于穩(wěn)態(tài)氙燈是高壓點(diǎn)亮,為了避免瞬間脈沖電流對(duì)周圍設(shè)備造成影響,需要先開氙燈,等電流穩(wěn)定后再打開周邊的電腦等設(shè)備。關(guān)機(jī)順序則相反。
2 軟件操作
熒光定量分析多采用工作曲線法,即以已知量的標(biāo)準(zhǔn)物質(zhì),按試樣相同方法處理后,配成一系列標(biāo)準(zhǔn)溶液,測(cè)定其相對(duì)熒光強(qiáng)度和空白溶液的相對(duì)熒光強(qiáng)度;扣除空白值后,以熒光強(qiáng)度為縱坐標(biāo),標(biāo)準(zhǔn)溶液濃度為橫坐標(biāo),繪制工作曲線;然后將處理后的試樣配成一定濃度的溶液,在同一條件下測(cè)定其相對(duì)熒光強(qiáng)度,扣除空白值后,從工作曲線上求出熒光物質(zhì)的含量。
在用熒光分析法測(cè)定某些會(huì)引起熒光改變的微量物質(zhì)時(shí),常采用差示熒光法,可使熒光強(qiáng)度的讀數(shù)差距拉大,使熒光強(qiáng)度的降低加大,從而提高方法的靈敏度和之。
